| I. Background | |
| CHRM5（Cholinergic Receptor Muscarinic 5）是毒蕈碱型乙酰胆碱受体（mAChRs）家 族成员之一，属于 GPCR，主要分布于中枢神经系统（如中脑多巴胺能神经元、纹状体） 及外周组织（如胰岛β细胞），顺利获得调控乙酰胆碱（ACh）信号影响奖赏通路、代谢稳态及 药物成瘾行为。ACh 结合 CHRM5 后激活 Gq/11 蛋白，激活 PLCβ通路，水解 PIP2 生成 IP 3 和 DAG，驱动钙离子释放及 PKC 活化；也顺利获得β-arrestin 通路调控 ERK 磷酸化，影响基 因表达与细胞增殖。 | |
| II. Description | |
| CHRM5 顺利获得 Gq 蛋白的信号通路，调控 IP1 的升高。CHRM5 与配体结合，导致 Gαq 亚基的激活，触发 PLC-γ，从而产生两个第二信使：IP3 和 DAG，它们参与细胞内钙动员。 由于 IP3 在代谢上不稳定，因此测量通路激活的替代标志物 IP1 作为检测标准。 CHRM5 CHO 细胞药靶模型很好的模拟了体内 CHRM5 的信号转导过程，原理见下图所示。  Figure 1. CHRM5 CHO 细胞模型原理图
